在一項(xiàng)新的研究中,來自瑞士日內(nèi)瓦大學(xué)、法國里昂大學(xué)和美國吉利德科技公司的研究人員發(fā)現(xiàn)我們的細(xì)胞如何自我抵御HBV感染,以及這種病毒如何反擊。這項(xiàng)研究代表著我們對(duì)HBV的理解取得重要進(jìn)展,并且提示著新的開發(fā)創(chuàng)新性治療試劑方法。相關(guān)研究結(jié)果于2016年3月16日在線發(fā)表在Nature期刊上,論文標(biāo)題為“hepatitis
B virus X protein identifies the Smc5/6 complex as a host restriction
factor”。

乙型肝炎病毒表面抗原(Hepatitis B Surface Antigen,
HBsAg)是位于乙型肝炎病毒(HBV)包膜表面的重要結(jié)構(gòu)蛋白,能誘導(dǎo)機(jī)體產(chǎn)生保護(hù)性免疫應(yīng)答。由HBsAg第124-147位氨基酸組成的“a”決定簇具有復(fù)雜的空間構(gòu)象,含有多個(gè)重要的免疫表位。發(fā)生在“a”決定簇或其周圍的變異影響HBsAg與抗體的結(jié)合,引起病毒的免疫逃逸。
來自南開大學(xué)生命科學(xué)學(xué)院的研究人員在新研究中證實(shí):乙型肝炎病毒X蛋白(hepatitis B virus X
protein,HBx)通過CREB介導(dǎo)YAP癌基因促進(jìn)了肝癌細(xì)胞生長。相關(guān)論文發(fā)表在12月的國際著名肝臟疾病雜志Hepatology上(影響因子11.665)上。
隨后,研究人員解析了HBx上調(diào)YAP的潛在分子機(jī)制。熒光素酶報(bào)告基因檢測(cè)揭示HBx對(duì)YAP nt
?232/+115這一包含cAMP反應(yīng)元件結(jié)合蛋白(CREB)元件的啟動(dòng)子區(qū)域進(jìn)行了調(diào)控。通過染色質(zhì)免疫沉淀法(ChIP),研究人員證實(shí)HBx能夠結(jié)合到Y(jié)AP的啟動(dòng)子上,但當(dāng)CREB沉默時(shí)它不能發(fā)揮作用。此外,借助于電泳遷移率實(shí)驗(yàn)(EMSA)和熒光素酶報(bào)告基因檢測(cè),他們也證實(shí)HBx激活了YAP啟動(dòng)子。在體內(nèi)外實(shí)驗(yàn)中,研究人員還證實(shí)YAP短干擾RNA(short
interfering RNA)能夠顯著阻斷HBx增進(jìn)肝癌細(xì)胞生長的效應(yīng)。
